Introduction: Several lines of the study showed that statins possess analgesics and anti-inflammatory properties. In present study. We examined the analgesic, anti-inflammatory and antioxidative effects of rosuvastatin in the experimental model of the chronic constriction injury model (CCI).
Methods: A current study designed in four groups (including Sham, CCI, CCI+ rosuvastatin (i.p. 5 mg/kg) and CCI + rosuvastatin (i.p. 10 mg/kg)). Heat hyperalgesia, cold and mechanical allodynia tests were performed on the 3th, 7th, 14th and 21st days after induced CCI. Finally, the blood samples were collected to measure the serum levels of TNF-α, and IL-6. Also, the spinal cord was collected to measure tissue concentration of MDA, SOD and GPx enzymes.
Results: The result of our study showed that CCI significantly increased heat hyperalgesia, cold and mechanical allodynia on 7th, 14th days and 21st days. Rosuvastatin attenuated CCI-induced hyperalgesia and allodynia.  Also, rosuvastatin treatment leads to reductions in TNF-α, IL-6, and MDA levels and increases in the concentration of SOD and GPx in CCI+Ros (5 mg/kg) and CCI+Ros (10 mg/kg) compared to CCI groups.
Conclusion: According to the results, attenuation of pain and inflammation in CCI-induced neuropathic pain by rosuvastatin treatment. It is suggested that antinociceptive effects elicited by rosuvastatin might be through the inhibition of inflammatory and antioxidant properties.
